A new nanofiber film developed by Cornell researchers offers a groundbreaking solution for those who struggle with swallowing pills. This innovative technology, detailed in the International Journal of Pharmaceutics, delivers a powerful combination of three drugs (acetylsalicylic acid, paracetamol, and caffeine) in an ultrafast-dissolving format, releasing 90% of the formulation in under two seconds. This rapid dissolution is a significant improvement over traditional tablets, which can take much longer to dissolve and release their active ingredients.

The key to this breakthrough lies in the choice of nanofiber material. Instead of opting for common edible polymers, the researchers selected cyclodextrin (hydroxypropyl-β cyclodextrin), a starch-derived cyclic oligosaccharide. This material has the unique ability to increase the water solubility of the drugs, enhance their bioavailability, and mask unpleasant tastes. The process of creating the nanofiber film is also environmentally friendly, utilizing water as the solvent rather than organic solvents, making it a "clean and green" approach.

The development of this film was a collaborative effort led by Tamer Uyar, an associate professor of fiber science, and Asli Celebioglu, a former postdoctoral researcher. Initially, the focus was on a single-drug formulation, but the team soon expanded to include a triple-drug combination, mirroring the composition of the commercial painkiller Excedrin. The challenge was to understand the complex interactions between the drugs and cyclodextrin, which they successfully achieved.

The results were remarkable. The nanofiber film demonstrated an incredibly fast dissolution rate, releasing 90% of the drug formulation in less than two seconds, a stark contrast to the powder formulation, which released only 30% in the same time. This discovery opens up exciting possibilities for the future of drug delivery, especially for those with difficulty swallowing pills. Uyar suggests that this proof of concept can be expanded to include dual or even quadruple drug combinations, further enhancing the effectiveness of pain relief.

The success of this project is attributed to the utilization of advanced facilities at Cornell, including the Center for Materials Research and the NMR Facility, supported by the National Science Foundation. This research not only addresses a practical issue for a specific population but also showcases the potential for nanofiber technology to revolutionize drug delivery, offering faster and more effective pain relief for a wide range of individuals.
